You have a problem with gnome somewhere. I would guess it is most likely in esd. Try stoping esd and start it from the command line. if you don't hear the startup tone than you have problems with esd. try as root and as user. if it is only users that cant play sound than you will need to add them to the audio group in /etc/group. I have only used midi along time ago in a galaxy far far away so I am not sure what to tell you for that, but I would guess that your permissions problem is the issue.
